Recombinant N-terminal fragment of parathyroid hormone (PTH 1-34) used to build bone in severe osteoporosis. Dosed once daily subcutaneously.
Mechanism of action
Intermittent PTH-receptor activation favors osteoblastic bone formation over resorption, raising bone mineral density.
Approved drug (Forteo). Serum half-life is ~1 h after SC dosing even though the biologic effect lasts longer. The former osteosarcoma boxed warning (rodent data) was removed in 2020.
